Betty made her cinematic début in a small, colourful role in the classic Clifton Webb comedy Sitting Pretty (1948). Following a minor role in Apartment for Peggy (1948), she received a big role as Barbara Bates’ younger sister in June Bride (1948), starring Bette Davis and Robert Montgomery. She continued the tradition by playing the bobbysoxer daughter of stars Loretta Young and Fred MacMurray in Mother Is a Freshman (1949) and Fred MacMurray and Maureen O’Hara in Father Was a Fullback (1950). (1950). After appearing in films starring Clifton Webb and Bette Davis (Cheaper by the Dozen (1950) and Payment on Demand (1951), respectively), her developing film career suffered significantly.

Betty changed her emphasis to 1950s television before returning to the stage for a rendition of “The Moon Is Blue” (1954). She continued to appear on the big screen from time to time, however she was largely uncredited or the films themselves were obscure. Betty auditioned for and was cast as Deputy Barney Fife’s steady gal on Andy Griffith’s homey sitcom while on leave from the Disney series “Texas John Slaughter” (1959). Knotts was a member of the small-town ensemble for five seasons before formally leaving the programme to pursue a full-time career in film. After Barney Fife left, Betty felt her role was incomplete without her link to Don Knotts, so the producers explored keeping her on the show.

She maintained her wholesome image on television with recurring roles on Family Affair (1966) and My Three Sons (1960), as well as guest appearances on The Farmer’s Daughter (1963), The Smith Family (1971), and Little House on the Prairie (1973). (1974). She was last seen in a Barnaby Jones (1973) episode in 1978, when she returned to the Mayberry cast for a reunion in the TV movie Return to Mayberry (1986).
